I separated survival association from experiments altering CMTM6. No raw-expression or mouse-data reanalysis was performed.\n\n**Findings.** Immunohistochemistry of **76 trastuzumab-treated HER2-positive tumors** associated high CMTM6 expression with poorer survival; these observational data do not show that inhibiting CMTM6 benefits patients. In resistant JIMT-1 cells, CMTM6 knockdown increased trastuzumab sensitivity; overexpression in SKBR3 cells had the opposite effect. Co-immunoprecipitation and ubiquitination assays support a HER2-stabilization mechanism. In xenografts, the figure describes **four mice per treatment group**; CMTM6-silenced tumors responded better to trastuzumab. These are cell-line-derived mouse tumors, not human treatment outcomes.\n\n**Limitations and uncertainty.** The clinical cohort is small and nonrandomized, while the intervention work uses engineered cells and small mouse groups.
